Apparently yes. Your speakers I will assume are square/rectangle in shape. You only need two of the four sides as reference (back and one side for example). In doing this I don't see why you would be having an issue placing, pushing, or sliding speakers over the tape.
If your hell bend on a laser pointer you simply need to put a reference point at/behind the listening position and use a standard position for the laser on the speaker. Easiest way would be the top using the tweeter as the center line. I suppose you could also make a simple jig to place the pointer in that will give your quick and easy placement.

Mount this to a simple base with a overhang on the front (think L shape with one short leg), mark center and your off to the races. You can pad the bottom with felt dots to protect speaker surfaces.
